Working on Optic Nerve Regeneration

Researchers are working on creating regeneration in mammals where it does not normally happen: "Researchers have long tried to get the optic nerve to regenerate when injured, with some success, but no one has been able to demonstrate recovery of vision. A team [now] reports a three-pronged intervention that not only got optic nerve fibers to grow the full length of the visual pathway (from retina to the visual areas of the brain), but also restored some basic elements of vision in live mice. ... [the mice were able to] regain some depth perception, the ability to detect overall movement of the visual field, and perceive light. ... Previous studies [have] demonstrated that optic nerve fibers can regenerate some distance through the optic nerve, but this is the first study to show that these fibers can be made to grow long enough to go from eye to brain, that they are wrapped in the conducting 'insulation' known as myelin, that they can navigate to the proper visual centers in the brain, and that they make connections (synapses) with other neurons, allowing visual circuits to re-form. ... [Researchers] combined three methods of activating the growth state of neurons in the retina, known as retinal ganglion cells: stimulating a growth-promoting compound called oncomodulin, [elevating] levels of cyclic adenosine monophosphate (cAMP) and deleting the gene that encodes the enzyme PTEN. ... these interventions have a synergistic effect on growth of optic nerve fibers. ... The eye turns out to be a feasible place to do gene therapy. The viruses used to introduce various genes into nerve cells mostly remain in the eye. Retinal ganglion cells are easily targetable."

Update in 07/2016: Since this post seems to have risen in the search engines and attracts a lot of interest from patients, passing readers may want to know that a different approach to stimulating retinal ganglion cell axons to regrow has achieved partial vision restoration in mice. It is an early and very qualified victory, and you should read the details carefully before becoming excited. Nonetheless, there is progress, albeit slow progress.

I totally believe the body is capable of healing itself; that this is what is most "natural." The research process described here fits...makes perfect sense. It sounds perfectly natural that nerve fibers would be capable of "...navigating to the proper visual centers...make connections with other neurons..." At a conference on spinal cord injury, I was once told that nerve fibers, once disconnected, would be difficult to be reconnected by doctors or scientists. I do not understand why it would not be natural that they find each other on their own (and properly re-connect).

He was told it's a side effect of whole brain radiation performed over a year ago. He is otherwise in great health and beating the cancer for 3 years. He is interested in any clinical trials or information regarding optic nerve regeneration via stem cells or otherwise. Any information is greatly appreciated.

My son age 35 years has lost sight from both of his eyes. At early age he was operated at Shankar netaralaya chennai and surgery was carried out. For some time it worked well but later again he landed into problems. 2nd time surgery was carried out at LV at Hyderabad but was not successful. Now he has no vision and he is totally dependent. The other complication are that he is not developed mentally fully. The point I carry all these years is whether he can be given eye sight. As per the report the nerve cells got calcification and are not in good health. If some body from eye surgeon fraternity can tell me whether in such cases any type of treatments are available in India.

Hi, I almost lost vision in my left eye (below 10/100) due to untreated glaucoma. I did not pay attention to very increased IOP (eye pressure) in my left eye until my vision has started to be blurred. Then doctors diagnosed glaucoma and I refused to undertake a classic surgery (laser surgery is inappropriate for my type of open-angle glaucoma). Paradoxically, I have managed to decrease IOP down to normal values (around 20) in my damaged eye by combination of eye drops and home-made eye drops based on natural ingredients (plants and hot/cayenne pepper). However, my vision cannot be restored unless my optic nerve damage is eventually repaired. Therefore, I am also very interested to participate in the trials. Thanks

My beautiful brave 9yr old daughter has an extremely rare bone disease in her skull, she has only just lost the sight in her right eye due to this. We were offered emergency de-compression surgery to take strain off the optic nerve but were informed it would not help her vision and the operation itself was alot for a 9yr old for the outcome to be the same in the end! Opthalmologist states she has healthy optic nerve and eye but just the slight compression to her nerve as caused her blindness. She has the bone disease around her left eye but not sure when she will loose the vision and trying out a drug to delay the abnormal bone growth compressing, the inevitable will happen in her left eye just don't know when as disease is so erratic. Please please please help with optic nerve regeneration, i can't speak for my daughter but i know she would want to try anything to at least keep one eye with vision....A child should never have to loose their eye sight, it must be the most scariest thing in the world, i know i am terrified for her and struggling to accept this.

My wife (67 years) underwent multiple surgeries e.g Radial Keratotomy resulting in Retinal detachment in both eyes which was successfully corrected by surgery. Subsequently underwent eye muscle surgery to fix double vision, followed by cataract surgery. She developed AMD in right eye which was fixed by single injection of AVASTIN. Six years ago she was diagnosed to have glaucoma. Her IOP could not be controlled well due to her being allergic to many eyedrops. She underwent Trabuctome surgery in one eye 9 months ago which made it worse.
In the past 6 months she kept loosing her vision and field sharply. Now she is legally blind with visual acuity 20/200 , 20/150. She has a Ahmed valve implanted in one eye 4 months ago which is able to control pressure. She is due for similar implant in the other eye in 2 weeks.
I wish her damaged optic nerves could be regenerated. We are devastated.

My Mother,46 year, having diabetes for three years, feels unable to read news paper lowlines about 6 months ago after removal of uterus due to fibroid, then she used spects of my father. About 2.5 months ago she feel slight 'dhundhlapan'(may be burl)in left eye only one time, apprx one month ago i have lost my left eye vision pain less within 10-15 days after it from 27/09/2015,, i have been given 5 steroids inj after then on oral steroids 60 mg withtaper by 20-20 mg for three days. after three days of steroid inj loss of vision in right eye with eyeache started and stopped at 3/60 within 4-5 days, when detereoration started in rt eye i have admitted in neurology AIIMS delhi where csf WAS NORMAL-pressure not more than 20, no cancer, NMO negative, mri orbit and brain -normal, with vision is 6/60 in right and 3/60 in left, they discharged us on continuation of 60 mg oral wysolone once a day with no cause found after taken the opinion of neuro opthalmolist who commented as 'No disc edema' on paper whille said to us as swelling in optic nurve. On repetion his comment was same on paper and said to us that swelling is as it is.
M'am after discharge, my mother feel 'dhundhlapan' (may be burl) in both eye which disappears in night. She had no vomiting,BP,headeche.
After 1 week, i ve gone to Neuro and opthal neuro in followup where vision was 6/36 in rt eye and 4/60 in left eye. but neurologist said us to followup in neuro opthal only with no comment on treatment that is being given by him.
neuro opthalmologist commented as swelling is as it is.
After it, we go for another neuro opthalmologist, She evaluated as complete disc pallor in left disc and coming in rt eye and said that it may optic neuritis. vision was 6/12 in right and 6/60 in left here. also she tapered the wysolone by 10-10 mg in three days.

presently my mother had no eyeache in left eye for last week and in right eye for two days. but saying improvement in vision in both eyes with burl.
when Dr neuro opthal evaluating, in last line, all the latters were readable exept first letter.and commented as 6/12 in rt and 6/60 in left.
also VER on 17/10/2015 is showing little abnormality with little high voltage in both eye.
what to do, is this pallor problem removable , is nurve recoverable . if not the will vision be stable.
is pallor controllable or may come/increse in rt eye.

After surgical removal of the tumor he experienced violent seizures, which have now stopped. He did regain some of his sight back. Not long after surgery he fell in his bathroom and lost all of his vision and has now been blind for four years. All who love him hate to see him suffer like this. Its mentally disturbing for him and all who love him. He has stated that he would participate in tests studies researches and procedures that would let him see again. His main desire is to see the faces of his children in any light stream or trickle of vision. Please contact me if our story makes him a candidate for any type of help. In addition this seems to be a possibly heridatary in his family. I don't think I have much of a right to post here, as many cases I have read are worse than mine and the people have suffered longer. Six weeks ago, I was a passenger who wasn't wearing a seatbelt in a vehicle being driven by someone I'd only met once before who was intoxicated and ran a red light. I don't remember leaving where we came from so I don't know what my head hit, as I broke my skull in two places and needed emergency surgery. Two small pieces of bone were lying against my left optic nerve. They didn't tear it but rubbed against it enough to lose 90-95% of my sight. I can't see light, even the sun, or tell if it's my mom or my son in front of me. I can see faint shapes of things and my sight is as if someone took a photograph and rubbed charcoal over it. Having only happened a short time ago, I'm still learning to be a one-eyed person...which is a blessing compared to some of the stories here and what COULD have happened to me in my accident. I hope there will quickly be progress made in the battle to restore peoples' eyesight. Good luck and best wishes to all who are praying for the same miracles.

My son's right eye vision was affected and when he did an MRI scan, we were shocked to be informed that a 3.7cm tumor compressed his optic nerve and damaged most of his right eye vision. He was 14 last year when he did his operation to remove the tumor. Later the doctor told us it was craniopharyngioma. But they can't revive his eye sight so he is left with one eye. Doctor said his right eye optic nerve is pale and pinkish which we pray and hope it may still be possible to revive the optic nerves.

My dad is old but he shouldn't have to live his remaining year blind and unhappy . His age should not be a factor in a study but a help to see if he can heal anyone should be able to. If a when a study is started I hope to add him to a list of people ready to get started.
I have a 5 year old grandson who I have just found out has a birth defect of his optic never also and can see light and dark only so this would be another good candidate at the other end of spectrum. Thank you

Hi,

Please tell the research progress on optic nerve regeneration, we are desperately looking for the positive outcome of this research so that my daughter can get back her vision which she lost due to meningitis in 2011 when she was 11 years old.
Please please include us also in tests or your research procedures.

Thanks

My 28 year old daughter has a benign optical nerve tumour that has affected both eyes as her optical nerves are damaged. We would be most interested to hear about trails, tests & research that is going on to give us hope that something can be done to improve or even better cure my daughters bad eye sight. Sie war im 7 Monat Schwanger und hat erst ihr Kind zur Welt gebracht per
Kaiserschnitt und am nächsten Tag war die Hirn OP.

Hello guys,

I recently went to the Dr. I was told that I have 90% optic nerve damage. I'm 30. And I've had glaucoma since the age of (i don't know) 12, perhaps. I don't know how long 10% of optic nerves will last me. But I am totally scared. Having read your stories, I've decided to pray not only for myself but for everyone here. All of your stories have touched me and I am 100% supportive of OUR journeys. We are embarking on an unknown. Guys, I'm frightened beyond words. But its comments and questions and stories like yours that makes me have the courage to face this unknown. Lord willing, and if He allows me to maintain vision with this bit that I have left, I promise you and Him that I will work overtime in donating and bringing people in on this cause. I am praying for you all. God bless, and just keep telling everyone you know about this. Thanks

My fiance suffered an optic nerve stroke in 2002 at the age of 21. In was/ is in terrific health. He is fully blind in his right eye since stroke. He is currently being treated for high pressure in eye reading at 38..he is on drops for glaucoma in hopes to decrease pressure ..also steroid drops and drops to help pain spasms. Due to corneal edema on top of issues they discussed today removing his eye Because of hemmorhaging that is not visible because of high pressure and blood settling. It's frustrating that eye removal is considered...we are desperate for another option as vision is not only concern. Always looking for information on further research of other options.

My son was born with healthy eyes. At the age of 4, he complained about his left eye.
He said he can only see objects from the side. So we went and made an appointment with an eye specialist for him. At that appointment the Dr said he needs to go under anaesthetic to do further investigation.
The results the Dr gave us are that he can't seem to find anything wrong. He said my son has got a lazy eye. So we tried all the methods the Dr said to improve the laziness. That December my son came to me and told me he can't see anything out of his left eye. It was close to Christmas and so we had to wait till January for all the Dr's to open. After the second appointment in his surgery he said my son has to go under another anaesthetic. A few days later we were in the hospital again for further investigation.
Results time came and the Dr said he can't be sure but he thinks it might be his optic nerve but can't seem to find something wrong with the eye. According to him my son is supposed to have full vision.
After that appointment we never went back because the Dr said there is nothing he can do for my son at that time. My son just turned 7years of age and is complaining that his right eye that has great vision is starting to hurt and complains about soreness in and around the eye while in school and doing homework.
We made an appointment with a new eye specialist for the 14th of August (earliest appointment they've got), but I am so scared to go through this again. My son is scared of hospitals and even if we come near one he starts to vomit.
Please I need help or options we could consider.
Many Thanks
Tammy

My husband has hereditary optic nerve atrophy. It was passed from his mother. One of his brothers had the same disorder. We have 4 children. Our youngest son that is 7 has recently been having trouble seeing. Everything is perfect on the surface but the optic nerves are showing signs that he may have what my husband his father has. The neuro ophthalmologist wants to put the knowledge of his dad's eye disorder to the side and is scheduling an mri to make sure there's not anything else going on such as something pushing on his optic nerves. He is leaning towards the possibility of it being the hereditary optic nerve atrophy since his dad has it but aim appreciative of his thoroughness to rule out any other causes. We are trying to wait patiently but would like to explore other options and find hope in treatments to restore his vision or at least stop him from losing any more. So possibly my son is a candidate for your trial research treatment. My husband would definitely be interested as well. We would love to be one of your success stories. Thanks for all that you do. You have our whole support in what you're doing to potentially better people's lives. May God bless you for that!

Hello!
My brother had a head injury that left him blind 10 years ago. The nerves in his Right eye were damaged and he lost his Right eyeball. The nerves and eyeball on Left side were intact. The doctors explained that he lost his vision because of the crossing over of the optic nerves at the optic chiasm. He has been entirely blind since that moment. We are interested in volunteering for trials and getting information on treatment options. Thank you so much for your help!
